Mechanism Of Action Of Sulfamethoxazole & Trimethoprim
- Sulfamethoxazole competitively inhibits dihydropteroate synthase, the enzyme responsible for bacterial dihydrofolic acid synthesis.
- Trimethoprim binds to dihydrofolate reductase and inhibits the reduction of dihydrofolic acid (DHF) to tetrahydrofolic acid (THF).
- Inhibition of this pathway prevents the synthesis of tetrahydrofolate and, ultimately, the synthesis of bacterial purines and DNA, resulting in a bacteriostatic effect.
- Tetrahydrofolate is an essential precursor in the thymidine synthesis pathway and interference with this pathway inhibits bacterial DNA synthesis.
